John McDuffie (1883-1950) was a federal judge on the United States District Court for the Southern District of Alabama.

He was nominated by President Franklin Delano Roosevelt on January 31, 1935, to replace Robert Tait Ervin. He was confirmed by the Senate on February 7th, and received commission on February 8th. He served until his death on November 1, 1950. McDuffie was succeeded in this position by Daniel Thomas.
